Tapentadol is a synthetic analgesic that acts centrally. It’s believed to work through two main mechanisms, although their clinical relevance is not entirely clear.
Tapentadol acts as a selective mu-opioid receptor (MOR) agonist. It binds to MOR with greater affinity than delta—and kappa-opioid receptors.
Additionally, Tapentadol inhibits noradrenaline reuptake. This action increases noradrenaline levels and activates alpha-2 receptors, promoting analgesia.
